چکیده
Enabling technologies in synthetic biology now present the opportunity to engineer wine yeast for enhanced novel aromas. In doing so, improved products will increase desirability of consumer and add value winemaker. The action enzyme carotenoid cleavage dioxygenase 1 (CCD1) on ?-carotene produce ?-ionone is interest improve aroma flavour wine. Engineering yeast, Saccharomyces cerevisiae, higher concentrations CCD1 grape-must presents an levels this volatile organic compound, thus enhancing organoleptic properties To end, four phylogenetically diverse plant genes were synthesised with a secretion signal peptide transformed into S. cerevisiae. relative ability each secreted supernatant cleave deep orange C40 was determined by spectrophotometry; furthermore, by-product such cleavage, highly aromatic C13 ?-ionone, assessed head-space solid-phase micro-extraction, analysis detection GCMS. Reduction release from validated LCMS CCD1. These experiments demonstrated that expression CCD1s derived Petunia hybrida Vitis vinifera their subsequent medium provided superior efficacy both reduction liberation. We anticipate knowledge being benefit future winemakers producing vinous product properties.

Production of β-ionone by combined expression of carotenogenic and plant CCD1 genes in Saccharomyces cerevisiae
BACKGROUND Apocarotenoids, like the C13-norisoprenoids, are natural compounds that contribute to the flavor and/or aroma of flowers and foods. They are produced in aromatic plants-like raspberries and roses-by the enzymatic cleavage of carotenes. Due to their pleasant aroma and flavour, apocarotenoids have high commercial value for the cosmetic and food industry, but currently their production ...
متن کاملCloning and functional characterization of carotenoid cleavage dioxygenase 4 genes
Although a number of plant carotenoid cleavage dioxygenase (CCD) genes have been functionally characterized in different plant species, little is known about the biochemical role and enzymatic activities of members of the subclass 4 (CCD4). To gain insight into their biological function, CCD4 genes were isolated from apple (Malus x domestica, MdCCD4), chrysanthemum (Chrysanthemum x morifolium, ...
متن کاملCAROTENOID CLEAVAGE DIOXYGENASE 7 modulates plant growth, reproduction, senescence, and determinate nodulation in the model legume Lotus japonicus
Strigolactones (SLs) are newly identified hormones that regulate multiple aspects of plant development, infection by parasitic weeds, and mutualistic symbiosis in the roots. In this study, the role of SLs was studied for the first time in the model plant Lotus japonicus using transgenic lines silenced for carotenoid cleavage dioxygenase 7 (LjCCD7), the orthologue of Arabidopsis More Axillary Gr...
متن کاملMAX3/CCD7 Is a Carotenoid Cleavage Dioxygenase Required for the Synthesis of a Novel Plant Signaling Molecule
BACKGROUND Plant development is exquisitely environmentally sensitive, with plant hormones acting as long-range signals that integrate developmental, genetic, and environmental inputs to regulate development. A good example of this is in the control of shoot branching, where wide variation in plant form can be generated in a single genotype in response to environmental and developmental cues. ...
